Dalam beberapa tahun terakhir, kemajuan signifikan telah dicapai dalam pemberian PROTAC secara selektif, yang berpotensi memperluas penggunaannya ke kondisi yang lebih luas (4).<\/p>\n<h2 class=\"pb-4 pt-2 text-2xl\"><strong>Bagaimana industri farmasi berinvestasi pada terapi penargetan proteasome?<\/strong><\/h2>\n<p class=\"\">Sejumlah terapi penargetan proteasome telah disetujui di Eropa dan Amerika Serikat, seperti Kyprolis (carfilzomib) dari Amgen, Ninlaro (ixazomib) dari Bristol Myers Squibb, dan Velcade (bortezomib) dari Johnson &#038; Johnson\/Takeda. Beberapa perusahaan Farmasi Besar\u2014AbbVie, AstraZeneca, Eli Lilly, Merck &#038; Co, Novartis, Pfizer, Roche, dan Sanofi\u2014memiliki produk dalam pengembangan klinis. Perusahaan bioteknologi AS lainnya yang bekerja di bidang ini termasuk Accutar Biotech, Cleave Therapeutics, Dialectic Therapeutics, dan Karyopharm Therapeutics.<\/p>\n<h2 class=\"pb-4 pt-2 text-2xl\"><strong>Apa sajakah pemain Eropa yang harus diwaspadai dalam bidang proteasome?<\/strong><\/h2>\n<p class=\"\">Beberapa bioteknologi Eropa sedang mengevaluasi kelas inhibitor baru yang memanfaatkan UPS, membajak proteasome untuk menghasilkan degradasi protein spesifik penyakit dengan berbagai aplikasi terapeutik mulai dari autoimun hingga onkologi.<\/p>\n<p class=\"\"><strong>Terapi Amphista (Inggris Raya) <\/strong>telah mengembangkan teknologi degradasi protein bertarget (TPD) generasi berikutnya untuk mengembangkan Lem Bertarget. Program utamanya, AMX-883, dapat menginduksi degradasi BRD9 untuk memblokir pertumbuhan tumor yang berasal dari pasien <em>hidup<\/em> sebagai monoterapi, dengan keunggulan dibandingkan venetoclax pada leukemia myeloid akut (AML) (13).<\/p>\n<p class=\"\"><strong>Terapi Booster (Jerman) <\/strong>berfokus pada pengembangan aktivator proteasome molekul kecil untuk penyakit neurodegeneratif menggunakan platform DGRADX-nya. Pada bulan Oktober 2025, perusahaan menyelesaikan putaran pembiayaan $15 juta yang dipimpin oleh investor ilmu hayati Apollo Health Ventures dan Novo Holding (14). Dana tersebut akan digunakan untuk memajukan jalur multi-penyakitnya.<\/p>\n<p class=\"\"><strong>OncoOne (Austria) <\/strong>berfokus pada pengembangan pengobatan presisi menggunakan terapi bertarget yang dibatasi lokasinya. Penelitian OncoOne mencakup pengembangan Imunoterapi bertarget yang memodulasi degradasi tumor menggunakan platform Pre-Targ-it. Perusahaan sedang menjajaki terapi yang menargetkan sumbu proteasome-Bcl-2 sebagai agen antikanker baru (15).<\/p>\n<p class=\"\"><strong>QLi5 Therapeutics GmbH (usaha patungan Jerman-Korea) <\/strong>sedang mengerjakan kelas baru dari proteasome inhibitor yang menunjukkan aktivitas anti-tumor dan anti-inflamasi yang kuat yang menjanjikan pada penyakit kanker, autoimun, dan inflamasi (16). Pada bulan September 2025, Qli5 menyelesaikan putaran pembiayaan Seri A senilai \u20ac10 juta (US$12 juta) dengan konsorsium investor internasional, termasuk SV Investment (Korea), KHAN Technology Transfer Fund I (Jerman), Atinum Investment (Korea), dan DAOL Investment (sebelumnya KTB, Korea). Hasil pendapatannya akan memajukan jalur inhibitor proteasome dalam pengembangan klinis (17).<\/p>\n<h2 class=\"pb-4 pt-2 text-2xl\"><strong>Peluang masa depan dalam menargetkan proteasome<\/strong><\/h2>\n<p class=\"\">Menurut riset pasar, pasar proteasome inhibitor global bernilai sekitar US$2,7 miliar pada tahun 2024 dan diproyeksikan mencapai US$3,03 miliar pada tahun 2024, tumbuh menjadi sekitar US$6,1 miliar pada tahun 2034, dengan tingkat pertumbuhan tahunan gabungan sebesar 8,70% dari tahun 2025 hingga 2034 (18). Penelitian awal menunjukkan bahwa proteasome mungkin memiliki efek yang luas dan dapat digunakan untuk menargetkan mekanisme pertahanan kekebalan baru (19).
